| Abstract: | Tato bakalářská práce obsahuje část teoretickou a praktickou. V teoretické části je popsán vznik, iniciace a růst nádorového onemocnění, včetně faktorů schopných vyvolat změny v genomu buněk organismu a genů, jejichž poškození vede k nádorové progresi. Dále je v teoretické části kapitola věnovaná metodám analýzy patogeneze nádorů, která zahrnuje jak hodnocení změn metodami molekulární biologie, ale i histologie a imunoanalýzy. Praktická část začíná definováním cílů a metodikou práce. Následuje část věnovaná metodám izolace NK a poté podrobný popis izolace NK na silikátovém povrchu, což je princip kolonkové metody, kterou jsem ve své práci použila. Následují naměřené parametry izolované DNA a RNA, statistické vyhodnocení a interpretace výsledků. |
| Abstract in different language: | This bachelor thesis consists of two parts: the theoretical one and the practical one. In the theoretical part there are described origin, initiation and growth of the neoplastic disease, including the factors triggering the changes in a genome of cells of organisms and genes, whose damage leads to a cancerous growth. Additionally, this part contains a chapter dedicated to the methods of analysis of tumor pathogenesis, which includes both evaluation of changes to the methods of molecular biology, but also histology and immuno-analysis. The practical part begins by defining the objectives and methodology of the work. The following is a section devoted to the methods of isolation of NA, and then a detailed description of the isolation of the NA above the surface, which is the principle column method, which I core in my work. The following are the measured parameters of the isolated DNA and RNA, statistical evaluation and interpretation of results. |
