| Abstract: | V bakalářské práci je porovnána referenční metoda průtoková cytometrie s výzkumnou metdou Pheno Immuno Computing. Teoretická část je zaměřena na popis zkoumaných T-buněk, jejich vývoj, populace a úlohu v imunitních procesech. Dále je zde uvedena charakteristika použitých metod. Praktická část je věnována ideálnímu postupu separace a kultivace buněk. Pro jejich následné měření PIC metodou byly zvoleny dva postupy popsané jako PIC short a PIC long. Vzorky pro průtokovou cytometrii byly zpracovaný jak pomocí referenčního postupu, tak pomocí experimentálního, jenž zahrnoval značení pomocí primární a sekundární protilátky. Pro zobrazení korelace bylo využito grafické zpracování. |
| Abstract in different language: | This bachelor thesis consists of comparing two methods. The referential method was flow cytometry, and it was compared with a new, experimental method: Pheno-Immuno Computing (PIC). The theoretical part of this thesis is focused on T-cells, their development, populations and their role in our immune system. Next chapters are about flow cytometry and PIC and their characteristics. The practical part consists of finding the ideal method for separating and cultivating the T cells, which are then being analyzed. Two forms of the PIC method were used PIC-short and PIC-long. Samples for flow cytometry were processed both in the referential and experimetnal way, using primary and secundary antibodies. Graphs were used to show the correlation between the two methods. |
